The film starts with the Super Troopers – Thorny (Jay Chandrasekhar), Mac (Steve Lemme), Foster (Paul Soter), and Rabbit (Erik Stolhanske) – all gathered at an Indian wedding celebration. The bride and groom are Thorny’s sister Sarita (Hannah Simone) and Farva (Kevin Heffernan). Rabbit asks how the hell they all got there.
Two months earlier, Sarita was driving toward Thorny’s place to crash. An obnoxious driver behind her honks and ends up ramming Sarita off the road and into the woods. Thorny overhears through her phone call and rushes to help her. Sarita’s car catches fire, but she is pulled out of the wreckage by Farva before the car explodes. The rest of the Troopers arrive and witness as Sarita instantly falls for Farva for saving her life. Needless to say, Thorny is extremely displeased.
The Troopers are now working under Captain Markowski (Nat Faxon) and have welcomed a new rookie, Coy Burns (Andrew Dismukes), while O’Hagan (Brian Cox) is retired. Markowski shows the guys a new electric patrol car, which they all think sucks. Meanwhile, Farva takes every chance he can get to rub it in Thorny’s face that he’s engaged to his sister. Also, Foster and Ursula (Marisa Coughlan) have broken up, though they still remain friends.
The guys pull up on the side to give Coy their list of pranks when they are spotted by brothers Steve and Baker Buchanan (Jon Rudnitsky and Chace Crawford), former pot dealers who now have their own party planning business. The Troopers previously busted them for vandalizing weed shops after it became legal and costing them money. The Buchanans taunt Thorny with Sarita and Farva’s wedding and then challenge the Troopers to a race to cross Duck Neck Bridge. Mac and Foster race the Buchanans, but their electric car’s safety features prevent them from gaining the necessary speed to make the jump, allowing the Buchanans to do so successfully.
Thorny decides that he has to stop the wedding when Farva keeps having sex with Sarita in Thorny’s house. Thorny arranges for his and Sarita’s parents, Drs. Makundan (Sakina Jeffrey and Iqbal Theba), to join them and Farva at a hotel for high tea so that they can see what Farva is really like. To Thorny’s surprise, Farva dresses up nicely and manages to charm Thorny and Sarita’s parents.
The Troopers learn that there is a new drug coming in from Canada called “Canadian Crystal”. Thorny responds to a suspicious package placed near a Moose Clock Syrup truck, which the locals hate because of “big syrup”. The Troopers bring a bomb robot to check out the package, but Foster becomes distracted at seeing Coy and Ursula appearing flirtatious, and he unintentionally detonates the C4 and causes a “syrup bukkake” explosion on everyone.
The guys join Thorny and his family for an Indian wedding celebration at a ranch, where the Buchanans and their cousin Bettie (Lisa Gilroy) are setting things up. Thorny gets annoyed by Farva’s behavior and organizes for them to do a discontinued ritual where Farva has to pick a snake from a basket. Thorny creates a distraction and shakes the basket to agitate the snakes. This makes one of them bite Farva in the neck. Thorny and Sarita’s mom Mehta sucks the venom out of his neck, causing not only Farva to get a boner, but also Mac, Foster, and Rabbit.
Thorny decides the ultimate way to break off the wedding is to have his dad witness Farva unleashed at his bachelor party. They go to a restaurant where Farva chases Mr. Makundan around and force-feeds him beef even though he is a vegetarian, but he ends up loving it. Markowski also ends up getting too high and drunk after accidentally eating Rabbit’s mushroom chocolate. Farva also makes the guys grab a ton of EpiPens because he is going to eat lobster despite being severely allergic to shellfish. Farva quickly turns red and starts running around madly, but the guys fling EpiPens at him until one of them actually injects him with one and brings Farva down.
Two strippers arrive for Farva, at which point Mr. Makundan believes that the wedding should be called off if Farva will be tempted by beautiful women. Thorny thinks he’s won and is about to get proof of Farva messing around, until he enters the next room and discovers his own dad having sex with one of the strippers. One of them faints and falls on Mr. Makundan’s leg, breaking it. After the ambulance arrives, Sarita finds out about Thorny’s scheming and disinvites him from the wedding. Markowski also ends up getting violently sick.
The Troopers go home for the night, even though Coy tries to keep the party going. Mac goes off to try and get the electric car to make the jump across Duck Neck Bridge, but the safety features take over and prevents him from doing so. Mac turns the car off and walks home but ends up coming upon the Buchanans and Bettie making their Canadian Crystal. Coy then shows up and knocks Mac out with a rifle.
Mac wakes up and learns that Steve, Baker, Bettie, and Coy are all in cahoots, as they make their drugs using Moose Clock Syrup. They set up the vandalism and were stealing the stuff themselves. They then put Mac in a large glass box and fill it with maple syrup to freeze him in it (like Han Solo). The electric car turns itself back on and crashes through the building to rescue Mac and drive to the wedding. The villains chase after them. The electric car then disregards its own safety features to jump Duck Neck Bridge.
Thorny is alone back at the station when he is visited by O’Hagan, who admits that he is bored with retirement. Thorny is trying to look over any footage to use against Farva, but he finds dashcam footage from the night of Sarita’s accident. It turns out Farva was the one who ran her off the road because he was dunking a hot dog in a Dr. Pepper and dropped it in there. Thorny thinks this will be what breaks them up for good.
Everyone gathers for the wedding. Thorny crashes it and gets ready to break the news, but Farva steps in and says he was inspired by Mr. Makundan admitting to his wife that he banged a stripper. He admits to Sarita that he ran her off the road but has genuinely fallen for her. Before she can respond, the electric car crashes through the venue. The guys use tea to melt the syrup off Mac’s mouth so that he can tell them about the Buchanans’ scheme and how Coy was in on it. He also mentions hearing that they are storing drugs in an elephant. Thorny thinks it’s an actual elephant and goes to stick his arm in the elephant’s ass to try and find drugs, but there is nothing in there. He realizes that there is an elephant statue in there, and he breaks it open, revealing the stash of Canadian Crystal. The Buchanans attempt to flee, but Steve and Baker are foiled, while Ursula blinds Bettie with piping hot samosas. Coy tries to take Sarita hostage, but Rabbit throws a knife into Coy’s chest.
The Buchanans are arrested. Mehta is thankful to Markowski after he took a bullet for her, and after seeing how sincere he is in his appreciation for Indian culture (the other Troopers tricked him into making him seem racist), she decides he will be her “revenge fuck” to make her and her husband even. Farva then tries to talk to Sarita again, who says that she is angry with him for lying to her and says that she now loves Rabbit because he was the one who saved her…but then it turns out she is just fucking with everyone and is ready to marry Farva.
Thorny ends up officiating not only Farva and Sarita’s wedding, but also Foster and Ursula, as well as Mac and the electric car (he is very high after being frozen in druggy maple syrup). Thorny begrudgingly accepts Farva as a brother-in-law, until he tells Thorny he’s going to be an uncle. Thorny sticks Farva in the neck with an Epi-Pen, leading to a series of antics that turn the rest of the ceremony into chaos.
During the credits, it’s shown that O’Hagan has been dating Governor Jessman (Lynda Carter) in his retirement. There is also a “tease” for “Super Troopers 4: The Hunt for O’Hagan’s Gold”.
